Is there any recomanded mysql tuning by sugar, for lots of small requests?

We are experiencing some performance issues with large customers. ~47 GB mysql database, ~140k requests/day, around half of which appear to be POST. There is no specifc pattern, so this is a generalized question. The mysql server is flooded with requests and at some point it lock the innodb resource down, and the other queries go into an infinite queue until server dies and is restarted automatically by linux.
