You can install Sugar Enterprise 10.1 and set your license key in the Instance. So it behaves like Sugar Serve. Or else if you want to setup in your local environment you can raise a ticket with Sugar Support Portal for requesting Instance Backup, So that Sugar Team will provide you the exact replica of your instance backup and with that you can set it up in your servers and you can use it for Development Purpose.

Levi Reeves10.1 is a cloud-only release, so there are not corresponding files in https://store.sugarcrm.com/download (assuming that's what you mean when you say "We have access to the build files"). 

The link that André Lopes sent is likely what you need as he pointed you to the Dev Builds group in SugarClub which stores the files we provide for customers looking to set up local testing environments for cloud-only releases (such as 10.1). However, your SugarClub user account is not associated with your company and therefore not verified that you should have access. If you follow the steps outlined here: https://support.sugarcrm.com/Resources/Setting_up_Access_to_the_Support_Portal/#Adding_Case_Portal_Access_to_Your_SugarCRMcom_Account, that should get your account associated with your company and therefore grant you access to the files.
